Building on the success of our original OliPure HIC technology, we've developed targeted purification protocols that address the unique requirements of specific oligonucleotide modifications.

Precision-Engineered for Fluorescent Probe Applications


The OliPure 6-FAM/BHQ-1 kit features optimized hydrophobic interaction chromatography specifically designed for dual-labeled probes synthesized on Kilobaser ONE XT systems. Our column chemistry employs silica-based support material functionalized with C18 alkyl chains, providing selective binding through hydrophobic interactions. The 300 Å pore size has been carefully selected to ensure optimal accessibility for oligonucleotide diffusion while maintaining high binding capacity.


This targeted approach recognizes that different oligonucleotide modifications require distinct purification strategies. By focusing on 6-FAM/BHQ-1 dual-labeled probes—widely used in qPCR, FISH, and other fluorescence-based applications—we've refined our protocols to deliver superior purity and yield for these specific constructs.


Streamlined Five-Step Protocol


The purification process remains remarkably straightforward:


  1. Binding phase: Crude probe dissolved in TEAA buffer (part of kit) binds to the hydrophobic resin
  2. First wash: 25% acetonitrile removes unbound 6-FAM and incomplete probes
  3. Salt removal: Aqueous wash eliminates residual salts and synthesis byproducts
  4. Elution: 50% acetonitrile releases the purified probe
  5. Dry: Use Kilobaser to dry the product

The entire process completes in under 10 minutes using standard laboratory centrifuges, with visual color changes providing immediate feedback on purification progress. The characteristic yellow eluent during the first wash confirms successful removal of excess fluorophore.


Enhanced Performance for Sensitive Applications


Our validation studies demonstrate that OliPure 6-FAM/BHQ-1 effectively removes the primary contaminants that interfere with fluorescence-based assays: unbound 6-FAM dye, salt residues from synthesis, and incomplete probes. This results in significantly reduced background fluorescence and improved signal-to-noise ratios in downstream applications.


The kit includes pre-measured solvents optimized for 6-FAM/BHQ-1 chemistry, eliminating buffer preparation and ensuring consistent results. Each kit provides materials for five purifications, with columns designed to handle the typical synthesis scales produced by Kilobaser ONE XT.
